+元々は[[三越]]や[[松下電器産業]]などの専属配送業者であったが、[[1976年]]に、当時の[[小倉昌男]]社長（故人）の決断により、個人を対象にした小口高速配送システム([[宅配便]])「[[宅急便]]」を開始して、大手企業からの専属配送事業から脱却を図る。

+近年は、企業からの発送である商流を主力とする[[佐川急便]]の取り扱い個数K伸びが著しく、集計方法によっては[[佐川急便]]がトップの場合もある。但し、商流の価格が安いため、依然ヤマト運輸が収益率は高い。

+ちなみに、ヤマト運輸も他の物流企業と同じく、通運業務・航空貨物輸送・引越し輸送業務を展開しているが、売上に占めるシェアは大きくない。

+かつて松下電器と関係があったためか、配達員が持っている端末はPanasonicである。また、三越店にては、荷受所を出店している。

+トヨタ自動車と宅配仕様バン型車「クイックデリバリー」を共同開発した縁からも、配送用小型トラック（市街地及び近郊配送用、最大積載量4t以下）及び１BOXバンの80％以上はトヨタ車である。

+郵政公社と対立はしているが、現段階では法律には勝てない為、日本郵政公社法に準じての配達のみ行うことができる。荷物の中に[[信書]]（手紙）を入れて送ることは信書便法に反する。ときたま荷物の中に手紙を入れて送る利用者がいるが、それを受けて配達をすると違法行為の一環となる。そのため、荷物の内容に関しては受け取り拒否される場合がある。更に、有価証券（商品券も含む）、現金などの類も禁止されている。宅急便に関しては30万円までの保証がされているが、上記禁止事項に違反している場合には、保証の対象とならない。
